Well let me start off with a note: the last time I used iTunes it used more CPU power and memory than either WinAmp or Media Player (the two biggest "competitors") which resulted in sound glitches and pops from time to time on my laptop. I steer well clear of iTunes for playing music - there's far better alternatives for that.
So, the logical question is: do you get these hiccups in Media Player or WinAmp (free download) too? If you don't, the answer is easy: iTunes hasn't been updated for Vista yet.
Vista uses a totally different and new system for handling sound - Apple is not a big Microsoft fan (for obvious reasons) so it wouldn't be surprising if they haven't added specific Vista improvements yet.
If other applications get the same sound glitch, it's most likely to the drivers - considering Vista is relatively new, it's not unexpected for certain drivers to not be perfect. Driver updates will be released though to improve Vista support - nearly all laptops use standard soundcards whose drivers are easy to find so no worries.
