Win16 subsystem error

Hello every one,

this is my first post over here, and thanks in advance for giving your time to analyse it. I use XP Pro, and i cant run a software anymore, which used to run before. Whenever i click the application a window comes up and says

System Error

The Win16 Subsystem was unable to enter Protected mode, DOS.EXE must be in your AUTOEXEC.NT and present in your path.
Its an old program called Jammer proffessional, but it used to run and god knows what happened now.

Can anybody help me on this? thanks again
