I cannot provide the answer why your method is not working, save that the link/executable is invalid.
I would fix the problem like so:
1) Remove the link from the Startup folder.
2) Goto Start Menu, Run, type: regedit
3) Browse to the following key:
HKEY_LOCAL_MACHINE->
SOFTWARE->
Microsoft->
Windows->
CurrentVersion->
Run
4) Right click the right hand pane of RegEdit (The values pane), right click some blank white space, select New->String Value.
5) Rename the String Value to whatever you wish (something relevant to the file you want to load).
6) Double click the new string value, and enter the path of the executable you wish to run on startup. Ensure you put long filenames in quotation marks: "C:\Program Files\Exe\MyExe.Exe"
