CHKDSK and defrag problem plz help

I am trying to defraq my computer and I am getting an erroe message saying Disk Defragmenter has detected that Chkdsk is scheduled to run on the volume: (C please run Chkdsk /f
Then i go to run Chkdsk /f and it says the type of file system is NTFS cannot lock the current drive. Chkdsk cannot run because the volume is in use by another process. Would you like to schedule this volume to be checked next time the system restarts? (Y/N) Y
Then when it restarts it automatically cancels CHKDSK. how do i fix this
