That actually sounds like a problem with the drive itself. You can force windows to refresh it's settings by simply removing the drive from the device manager then restarting the system. It should re-install the drive and reset all of the stored settings for that drive, such as autoplay.
If that doesn't work it may just be the drive. They do go bad sometimes. To test this you can try to boot from a cd such as the windows xp install disc, if it won't even do that then i'd suggest trying another drive.
